Abstract
A method was developed to enhance the carbohydrate homogeneity of recombinant protein. The secreted protein was purified using a combination of ultrafiltration, lectin affinity chromatography, ion exchange chromatography and reverse-phase HPLC. The disulphide linkages and the glycoform distribution on all three glycosylation sites were characterized by combining N-terminal sequencing with matrix-assisted laser desorption ionization time of flight (MALDI-TOF) and ESI FT-ICR MS. The techniques confirmed the expected disulphide linkage pattern.
