Abstract

The determination of hydrogen peroxide and peroxynitrite-induced modifications to rabbit skeletal troponin C by online liquid chromatography electrospray Fourier transform ion cyclotron resonance mass spectrometry (LC ESI FT-ICR MS) was discussed. Muscular weakness resulted from modifications to EF- hand proteins in the contractile apparatus of muscle fibers. Methionine residues 157 and 158 were susceptible to oxidation and contributed to loss of function in TnC. Protein modifications were mapped to specific residues by comparing tryptic digests of treated vs. control TnC. It was found that the data was analyzed with MIDAS software.
